DEFINITION
3-[(4-Amino-2-methylpyrimidin-5-yl)methyl]-5-(2-hydroxyethyl)-4-methyl-1,3-thiazol-3-ium chloride hydrochloride.
Content: 98.5 per cent to 101.0 per cent (anhydrous substance).
CHARACTERS
Appearance: White or almost white, crystalline powder or colourless crystals.
Solubility: Freely soluble in water, soluble in glycerol, slightly soluble in ethanol (96 per cent), practically insoluble in heptane.

A. Infrared absorption spectrophotometry.
B. Dissolve about 20 mg in 10 mL of water, add 1 mL of dilute acetic acid and 1.6 mL of 1 M sodium hydroxide, heat on a water-bath for 30 min and allow to cool. Add 5 mL of dilute sodium hydroxide solution, 10 mL of potassium ferricyanide solution and 10 mL of butanol and shake vigorously for 2 min. The upper alcoholic layer shows an intense light-blue fluorescence, especially in ultraviolet light at 365 nm. Repeat the test using 0.9 mL of 1 M sodium hydroxide and 0.1 g of anhydrous sodium sulfite instead of 1.6 mL of 1 M sodium hydroxide. Practically no fluorescence is seen.
C. It gives reaction of chlorides.
TESTS
Solution S: Dissolve 2.5 g in carbon dioxide-free water and dilute to 25 mL with the same solvent.
Appearance of solution: The solution is clear and not more intensely coloured than reference solution
pH: 2.7 to 3.3.

DESCRIPTION
Thiamine Hydrochloride occurs as small, white to yellow-white crystals or crystalline powder. When exposed to air, the anhydrous product rapidly absorbs about 4% of water. It melts at about 248C with some decomposition. One-gram dissolves in about 1 mL of water and in about 100 mL of alcohol. It is soluble in glycerin and is insoluble in ether.
Function: Nutrient.
Identification
A. The infrared absorption spectrum of a potassium bromide dispersion of the sample, previously dried at 105C for 2 h, exhibits relative maxima at the same wavelengths as those of a similar preparation of USP Thiamine Hydrochloride Reference Standard.
B. A 1:50 aqueous solution gives positive tests for Chloride,
Color of Solution: Passes test.
Lead: Not more than 2 mg/kg.
Nitrate: Passes test.
pH of a 1:100 Solution: Between 2.7 and 3.4.
Residue on Ignition: Not more than 0.2%.
Water: Not more than 5.0%.
