DEFINITION
Ammonium Glycyrrhizate is a mixture of ammonium 18α- and 18β-glycyrrhizate (ammonium salt of (20β)-3β-[[2-O-(β-d-glucopyranosyluronic acid)-α-d-glucopyranosyluronic acid]oxy]-11-oxoolean-12-en-29-oic acid), and the 18β-isomer is the main component. It contains NLT 78.0% and NMT 102.0% of ammonium 18α- and 18β-glycyrrhizate, on the anhydrous basis.
IDENTIFICATION
A. The retention times of the peaks of 18α- and 18β-glycyrrhizic acid from the Sample solution correspond to those from the System suitability solution, as obtained in the Content of Ammonium 18α- and 18β-Glycyrrhizate. [Note— The peak of 18α-glycyrrhizic acid could be absent in the Sample solution.]
B. Identification Tests—General, Ammonium.

Acceptance criteria: NMT 0.5%
Limit of Organic Impurities: To pass the test by chromatography.
Optical Rotation, Specific Rotation:
Sample solution: 10.0 mg/mL of Ammonium Glycyrrhizate in 50% ethanol
Acceptance criteria: +49.0 to +55.0 on the anhydrous basis
Water Determination: Acceptance criteria: NMT 6.0%

DEFINITION
Mixture of ammonium 18α- and 18β-glycyrrhizate (ammonium salt of (20β)-3β-[[2-O-(β-D-glucopyranosyluronic acid)-α-D-glucopyranosyluronic acid]oxy]-11-oxoolean-12-en-29-oic acid), the 18β-isomer being the main component.
Content: 98.0 per cent to 102.0 per cent (anhydrous substance).
CHARACTERS
Appearance: White or yellowish-white, hygroscopic powder.
Solubility: Slightly soluble in water, very slightly soluble in anhydrous ethanol, practically insoluble in acetone. It dissolves in dilute solutions of acids and of alkali hydroxides.
IDENTIFICATION
A. Infrared absorption spectrophotometry
B. Dissolve 0.1 g in 20 mL of water, add 2 mL of dilute sodium hydroxide solution R and heat cautiously. On heating, the solution gives off vapours that may be identified by the alkaline reaction of wet litmus paper.
Appearance of solution: The solution is clear and not more intensely coloured than reference solution.
Dissolve 1.0 g in ethanol (20 per cent V/V) and dilute to 100.0 mL with the same solvent.
Specific optical rotation: + 49.0 to + 54.0 (anhydrous substance).
Dissolve 0.5 g in ethanol (50 per cent V/V) and dilute to 50.0 mL with the same solvent.
Related substances: To pass the test by Liquid chromatography.
Water: Maximum 6.0 per cent, determined on 0.250 g.
Sulfated ash: Maximum 0.2 per cent, determined on 1.0 g.
Storage: In an airtight container.
